I am in Canada, on a Work Permit and would like to apply for PR. Which is the best & fastest way to do this.
Should I apply for PR with CIC directly or should I go through PNP and then apply to CIC.

I would expect a standard Federal skilled worker application to be the best route in your case, assuming that you are not working in the province of Quebec and/or do not intend to settle in that province. This is somewhat speculative, however, without all the information relevant to the case.
