Your options are to pursue an inland sponsorship, or to have the application processed at a visa office abroad. In the context of an inland sponsorship, you can seek an open work permit after approval-in-principle. As this can take as many as a few years to obtain, you may need to seek a work permit to be able to remain in your present employ upon expiration of your current visa.

With my Job Offer and LMO can I apply at any Canadian Port of Entry? I am an Australian Citizen, CIC website indicates the following:

Applying for a Work Permit at the Canadian Border
You can only apply for a permit when you arrive in Canada if:

You do not need a medical examination.
You are from theUnited States, Greenland or Saint-Pierre and Miquelon.
You do not need a temporary resident visa (TRV) to visit Canada and your job does not need a labour market opinion from Human Resources and Social Development Canada (HRSDC).
You do not need a temporary resident visa to visit Canada, your job requires you to have an HRSDC labour market opinion and it has been issued by the time you arrive.
